Manesso E, Toffolo GM, Saisho Y, Butler AE, Matveyenko AV, Cobelli C, Butler PC. Dynamics of -cell turnover: evidence for -cell turnover and regeneration from sources of -cells other than -cell replication in the HIP rat. Am J Physiol Endocrinol Metab 297: E323–E330, 2009. First published May 26, 2009; doi:10.1152/ajpendo.00284.2009.—Type 2 diabetes is characterized by hyperglycemia, a deficit in -cells, increased -cell apoptosis, and islet amyloid derived from islet amyloid polypeptide (IAPP). These characteristics are recapitulated in the human IAPP transgenic (HIP) rat. We developed a mathematical model to quantify -cell turnover and applied it to nondiabetic wild type (WT) vs. HIP rats from age 2 days to 10 mo to establish 1) whether -cell formation is derived exclusively from -cell replication, or whether other sources of -cells (OSB) are present, and 2) to what extent, if any, there is attempted -cell regeneration in the HIP rat and if this is through -cell replication or OSB. We conclude that formation and maintenance of adult -cells depends largely ( 80%) on formation of -cells independent from -cell duplication. Moreover, this source adaptively increases in the HIP rat, implying attempted -cell regeneration that substantially slows loss of -cell mass.

Augmenting Peripheral Nerve Regeneration Using Rat Hair Follicle Stem Cells (rHFSCs) in Rats

Introduction: Nowadays, cell therapy is the most advanced treatment of peripheral nerve injury. The aim of this study was to determine the effects of transplantation of hair follicle stem cells on the regeneration of the sciatic nerve injury in rats. Methods: The bulge region of the rat whisker was isolated and cultured. Morphological and biological features of the cultured bulge cells were ob...

Facts about Stem Cells and Importance of Them

Stem cells are undifferentiated biological cells that can differentiate into specialized cells and can divide (through mitosis) to produce more stem cells. They are found in multicellular organisms. In mammals, there are two broad types of stem cells: embryonic stem cells, which are isolated from the inner cell mass of blastocysts, and adult stem cells, which are found in various tissues. In ad...

Tooth Regeneration with Stem Cell Sources

Introduction: During the last decade, advances in tissue engineering and stem cell-based tooth regeneration have provided realistic and attractive means of replacing lost or damaged teeth. The first adult stem cells isolated from dental tissues were dental pulp stem cells (DPSCs). When transplanted with hydroxyl apatite/tri calcium phosphate (HA/TCP) powder, they formed a dentin-like structure...

Co-Culture of Mesenchymal Stem Cells with Mature Chondrocytes: Producing Cartilage Construct for Application in Cartilage Regeneration

Background: Cell-based treatment approach using differentiated mesenchymal stem cells (MSCs) and mature chondrocytes has been considered as an advanced treatment for cartilage repair. We investigated the differentiated level of these two cell types that is crucial for their repair capacity for cartilage defect at a co-culture micro mass system. Methods: Passaged-2 MSCs isolated from the mouse b...

Mesenchymal Stem Cells as an Alternative for Schwann Cells in Rat Spinal Cord Injury

Background: Spinal cord has a limited capacity to repair therefore, medical interventions are necessary for treatment of injuries. Transplantation of Schwann cells has shown a great promising result for spinal cord injury (SCI). However, harvesting Schwann cell has been limited due to donor morbidity and limited expansion capacity. Furthermore, accessible sources such as bone marrow stem cells ...
